Materials of Disintegrant

Process of Disintegrant

3. Blending: The API is blended with the disintegrant and other excipients (such as binders, lubricants, and fillers) in a suitable blender to ensure uniform mixing. The proportion of disintegrant is critical and is determined during the formulation development stage.
4. Granulation (if necessary): If the powder blend is not directly compressible, a granulation process may be used. This involves adding a liquid binder to the powder blend to create granules that are easier to compress. The granules are then dried and milled if necessary.
5. Tablet Compression: The blended or granulated material is loaded into a tablet press, where it is compressed into tablets under high pressure. The disintegrant is evenly distributed throughout the tablet to ensure even disintegration.
6. Drying and Finishing: After compression, the tablets may need to be dried to remove any remaining moisture. They are then finished by processes such as coating (if required) and are polished to ensure a smooth surface.
7. Quality Control Testing: The tablets are tested for various parameters, including disintegration time, to ensure they meet the specified quality standards. Disintegration testing is typically done using a disintegration test apparatus, where the tablet is subjected to simulated gastrointestinal fluids to measure the time it takes to disintegrate.

How to improve disintegration of tablets?
Disintegrants are composed of various components that work together to facilitate the rapid disintegration of tablets in the gastrointestinal tract, including natural polymers like modified starches and cellulose derivatives, synthetic polymers such as crosslinked polyvinylpyrrolidone and graft copolymers, inorganic compounds like bentonite and magnesium aluminometasilicate, natural product-based disintegrants from sources such as marine algae and plant cell walls, ionic exchange resins, chemically modified natural products, and hybrid mixtures of these materials, all of which are selected and combined to optimize the tablet's disintegration performance.

How to improve disintegration of tablets?
In general, an increase in particle size improves the disintegration and dissolution of tablets. This attributes to the efficient formation of hydrophilic networks by larger size particles. Degree of substitution relates to the total acidic (acid form) and basic (sodium salt) components of superdisintegrants.

At the end of the year, polyvinylpyrrolidone K30 was produced, and the quality reached Industrial grade, some of which have reached European standard pharmaceutical grade, and are being tested by users and SGS and other professional testing institutions; now PVPK90 products are in trial production. The project currently has completed fixed assets of approximately 110 million yuan.
